Melanoma mutations modify melanocyte dynamics in co-culture with keratinocytes or fibroblasts

Abstract: Melanocytic cell interactions are integral to skin homeostasis and affect the outcome of multiple diseases, including cutaneous pigmentation disorders and melanoma. Using automated-microscopy and machine-learning-assisted morphology analysis of primary human melanocytes in co-culture, we performed combinatorial interrogation of melanocyte genotypic variants and functional assessment of lentivirus-introduced mutations. Keratinocyte-induced melanocyte dendricity, indicator of melanocyte differentiation, was redu… Show more

“…Cells were stained with anti-dsDNA Ab (Merck) and developed with the appropriate fluorescently labelled secondary antibody. Plates were imaged in an InCell Analyzer 2000 (Cytiva, North Ryde, NSW, Australia) and the cytoplasmic pixel intensity analysed as previously [23]. To validate the specificity of the dsDNA Ab staining, fixed cells were treated with DNAse1 (Roche) following the permeabilisation step by adding 200 U per well and incubating at 37 • C for 30 min before washing in PBS and proceeding with the blocking step.…”

“…Indeed, melanocytes display differentiated “dendritic shapes” in healthy skin, differing from melanoma cell morphology. This differentiated morphology has been largely described for melanoma cells, namely those treated with antitumor agents [ 20 , 21 , 22 ].…”

“…This interaction is more pronounced in response to UV exposure. We have also seen the ability of keratinocytes to modify melanoma cell morphology in vitro [ 17 ] and decrease Tyr expression, a key player in pigmentation and the control of the Ink4A-Arf locus of genes [ 18 ]. Based on these observations, we hypothesized that epidermal keratinocytes impact OIS in melanocytes.…”
