Mefunidone Attenuates Tubulointerstitial Fibrosis in a Rat Model of Unilateral Ureteral Obstruction

Abstract: BackgroundInflammation has a crucial role in renal interstitial fibrosis, which is the common pathway of chronic kidney diseases. Mefunidone (MFD) is a new compound which could effectively inhibit the proliferation of renal fibroblasts in vitro. However, the overall effect of Mefunidone in renal fibrosis remains unknown.MethodsSprague-Dawley rats were randomly divided intro 6 groups: sham operation, unilateral ureteral obstruction (UUO), UUO/Mefunidone (25, 50, 100mg/kg/day) and UUO/PFD (500mg/kg/day). The rat… Show more

“…We also investigated the accumulation of type I and III collagen, which are major components of the extracellular matrix that accumulate in renal fibrosis. The kidneys in the Control group but not Sham group exhibited a significant increase in the deposition of both types of collagen on days 7 and 14, consistent with other studies . Notably, Thalido treatment reduced the deposition of both types of collagen on days 7 and 14.…”

“…Immunohistochemistry (IHC) staining was performed in paraffin sections as previously reported [29]. Slides were incubated with a primary antibody against collagen I (1:500), collagen III (1:800), CD3 (1:200), CD68 (1:200), or CBS (1:500).…”

“…To repair and regenerate tubular function after acute renal disease, HGF is a key protein [36,37,39,40] which is not only considered as an endogenous anti-fibrotic factor, but also as therapeutic element that blocks renal tissue fibrosis and repair dysfunction [44]. In addition, HGF prevents accumulation of collagen in renal fibrosis [45,46]. Fig.…”
