Media development for large scale Agrobacterium tumefaciens culture

Abstract: A chemically defined media was developed for growing Agrobacterium tumefaciens at large scale for commercial production of recombinant proteins by transient expression in plants. Design of experiments was used to identify major and secondary effects of ten media components: sucrose, ammonium sulfate ((NH ) SO ), magnesium sulfate heptahydrate (MgSO *7H O), calcium chloride dihydrate (CaCl *2H O), iron (II) sulfate heptahydrate (FeSO *7H O), manganese (II) sulfate monohydrate (MnSO *H O), zinc sulfate heptahydr… Show more

“…The vitality of A. tumefaciens was found to be in uenced by the bacterium culture medium, growth period, and environmental conditions. In this study, we utilized YEP medium for culturing A. tumefaciens, as it is known to be particularly suitable for A. tumefaciens cultivation 47 . After an overnight recovery in the shaker, the EHA105 strain demonstrated robust vitality with rapid multiplication rates.…”

“…Indeed, the ambiguity of the composition of LB leads to ambiguity in the spectroscopy and the interpretability of the behavior of the various contributors to the IE. This situation prevails in spite of a strong shift in research and applied bacteriology [25][26][27][28][29][30] toward use of so-called ''chemically defined'' media (CD), of which minimal medium (MM) is an example.…”

“…CD media provide improved control and reproducibility of the cultures. [25][26][27][28][29][30] LB media contain a broad range of materials, such as proteins, carbohydrates, and lipids as relatively unrefined biological sources, e.g., yeast extract or trypsinized milk. They are thus complex mixtures that defy the detailed analysis that is possible with CD media.…”

“…33 In a study in which a chemically defined media was developed for growing Agrobacterium tumefaciens at large scale for commercial production of recombinant proteins by transient expression in plants, PBD was employed. 35 On the other hand, in recent times, researchers have been modeling the results of their own fermentation studies. The maximum specific growth rate of Agrobacterium tumefaciens in a chemically defined media was 0.33 hr −1 and the final biomass concentration was found as 2.6 g dry cell weight/L.…”
