Mechanomics Approaches to Understand Cell Behavior in Context of Tissue Neogenesis, During Prenatal Development and Postnatal Healing

Abstract: Putra et al. Live Imaging Mechanomics Tools approaches and methodologies implemented to date in a series of studies carried out by our consortium. The consortium's body of work is described in context of current roadblocks in the field and innovative, breakthrough solutions and is designed to encourage discourse and cross disciplinary collaboration in the scientific community.

“…Broadly speaking, mechanomics is a study of omics data within the context of mechanobiology. So far, this term was used with regard to changes in omics profiles in response to a mechanical stimuli such as shear flow, tensile stretch, or mechanical compression [33][34][35] , or to collectively name all of the mechanical forces acting on or within cells [36][37][38][39][40] . However, it can also be used to address omics changes related to changes in the mechanical properties of cells 41,42 -a context much closer to our study.…”

“…Throughout prenatal development as well as during postnatal healing, which repeats prenatal development processes, cells interconvert between epithelial and mesenchymal states (EMTs and EMTs, as per above), enabling the formation of complex geometries, tissue architectures, and organ systems ( Figures 1A,B ; Morali et al, 2004 ; Putra et al, 2020 ). The process is so ubiquitous that the formation of deleterious tissues such as cancerous tumors and metastasis of circulating tumor cells follow analogous paradigms ( Jolly et al, 2017 ).…”

“…The manuscript proposes a paradigm shifting approach to understand the cellular underpinnings of diseases as different as osteoarthritis and early onset dementia in bone and brain. We know as biologists that cells manufacture, remodel and adapt tissues throughout life ( Knothe Tate et al, 2016a ; Putra et al, 2019 ). The tissues render physically the collective cellular experience, reflected in architectures (bones) and memories (brain) which themselves exhibit emergent properties ( Knothe Tate, 2020 ).…”
