Mechanobiology of MG63 Osteoblast-Like Cells Adaptation to Static Magnetic Forces

Abstract: The aim of this study was to explore the biophysical effects of static magnetic field on osteoblastic cells. MG63 cells were exposed to 0.25 and 0.4-T static magnetic fields (SMF). The cell cycle effects were tested by flow cytometry. The differentiation of the cells was assessed by detecting the changes in prostaglandin E2, osteocalcin, and extracellular matrix expression. Membrane fluidity was used to evaluate the alterations in the biophysical properties of cellular membranes after the SMF simulations. Our … Show more

“…The mRNA expressions were analyzed by RT-PCR (a) and the band intensities were further quantified (b). *, p \ 0.05 (n = 3; compared the difference between SMF-exposed and unexposed groups in basal or osteogenic culture medium) (Con) control, unexposed group; (SMF) static magnetic field; (Basal) basal medium; (Dex/b-GP) dexamethasone/ b-glycerophosphate exposure did not cause DNA damage or affect cell viability (Lin et al 2008;Amara et al 2007). AlliotLicht et al (2005), however, indicated that Dex significantly inhibited the proliferation of human DPCs but conversely stimulated ALP activity in a 7-day culture.…”

“…In addition, several recent studies provide evidence that SMF exposure infl uences the cell membrane structure (Higashi et al 1993, Tiburu et al 2001, Chiu et al 2007, Lin et al 2008). However, no study provided information regarding the combined eff ects of SMF exposure and the slow freezing process on human erythrocytes.…”

“…A higher value of fl uorescence anisotropy can refl ect lower membrane fl uidity and higher structural order of the cell membrane (Vest et al 2006, Marczak 2009). Several researchers found that MG63 cells exposed at 0.4 T had signifi cantly increased cell membrane rigidity, with an increased related ratio of about 15% (Chiu et al 2007, Lin et al 2008). In addition, their results demonstrated that the major alteration in fl uorescence anisotropy was in the hydrophilic region of the phospholipid bilayer, but not in the hydrophobic region (Lin et al 2008).…”
