Mechanisms Regulating Abnormal Circular RNA Biogenesis in Cancer

Abstract: Circular RNAs (circRNAs), which are a class of endogenous RNA with covalently closed loops, play important roles in epigenetic regulation of gene expression at both the transcriptional and post-transcriptional level. Accumulating evidence demonstrated that numerous circRNAs were abnormally expressed in tumors and their dysregulation was involved in the tumorigenesis and metastasis of cancer. Although the functional mechanisms of many circRNAs have been revealed, how circRNAs are dysregulated in cancer remains … Show more

“…The circRNAs play critical biological functions in eukaryotic organisms, which compete for miRNAs. By base-complementary pairing with the target mRNA 3-UTR, miRNAs can block the translation and stability of target RNA-binding Proteins (RBPs) can interact with circRNAs and regulate circRNA splicing, replication, folding, stabilization and localization ( Huang and Zhu 2021 ; Zeng et al, 2021 ). In summary, the circRNAs act as miRNA sponges and interact with RBPs to perform transcriptional functions in organisms.…”

“…Circular RNAs (circRNAs) are covalently contiguous closed loops without 5′ and 3′ ends, and are structurally more stable than linear RNAs and less susceptible to degradation by nucleic acid exonucleases ( Jakobi and Dieterich, 2019 ; Huang and Zhu, 2021 ). Initially circRNAs were thought to be products of missplicing or intermediates escaping from the lasso structure of introns ( Chen and Huang, 2018 ; Tsitsipatis and Gorospe, 2021 ).…”

“…Circular RNAs (circRNAs)-a particular type of endogenous noncoding RNA-were first identified in viruses in the 1990s (8). Initially, circRNAs were considered to be the product of missplicing.…”
