Mechanisms of Autoantibody Production in Systemic Lupus Erythematosus

Abstract: Autoantibodies against a panoply of self-antigens are seen in systemic lupus erythematosus, but only a few (anti-Sm/RNP, anti-Ro/La, anti-dsDNA) are common. The common lupus autoantigens are nucleic acid complexes and levels of autoantibodies can be extraordinarily high. We explore why that is the case. Lupus is associated with impaired central or peripheral B-cell tolerance and increased circulating autoreactive B cells. However, terminal differentiation is necessary for autoantibody production. Nucleic acid … Show more

“…Several of these autoantigens therefore have a nuclear localization [21]. Below is an exploration of the main target antigens in the ACTD and a brief explanation of their roles and functions along the pathway of protein synthesis and secretion, herein described as the transcription, translation, transport-trail (TTTT or 3T-trail) [22,23].…”

“…The rapid-onset disease in males is attributable to the Y-autoimmune accelerator ( Yaa ) locus, which is due to a translocation from the X to the Y chromosome, duplicates 16 genes, including TLR7 (29, 30). TLR7, regulates the activation of the type 1 IFN pathway by ribonucleic acid complexes, a critical pathway in SLE pathogenesis (31). Therefore, in spite of its presentation in males, the BXSB/ Yaa strain is uniquely suited to model the consequences of an overeactive TLR7/Type 1 IFN pathway.…”

W przebiegu TRU jest wytwarzana szeroka gama przeciwciał skierowanych przeciwko własnym antygenom, jednakże tylko kilka z nich można uznać za typowe dla tej choroby, tj.…”
