Mechanism of the circadian clock in physiology

Abstract: It has been well established that the circadian clock plays a crucial role in the regulation of almost every physiological process. It also plays a critical role in pathophysiological states including those of obesity and diabetes. Recent evidence has highlighted the potential for targeting the circadian clock as a potential drug target. New studies have also demonstrated the existence of "clock-independent effects" of the circadian proteins, leading to exciting new avenues of research in the circadian clock f… Show more

“…Now it is well established that circadian rhythms exist in almost all life forms, from simple archaebacteria to complex humans [2]. In mammals, mounting evidence has demonstrated that the circadian clock regulates a multitude of physiological functions including but not limited to: metabolism, immune response, renal function, sleep-wake cycles, and blood pressure [3, 4]. Interestingly, it has been shown that deregulation of the circadian clock can also be attributed to certain pathological states, including hypertension [5].…”

“…THE CIRCADIAN CLOCK IS AN important regulator of many physiological functions including blood pressure; metabolism; and endocrine, immune, vascular, and renal function (1,6,30,40,43,44,49). The canonical circadian clock molecular mechanism, known as the transcription translational oscillating (TTO) loop, involves four core proteins that interact with each other to regulate transcription of circadian target genes (3).…”

“…The decline in BP during sleep may be attributed in part to the Zeitgeber factors that reduce blood pressure and natriuresis by various molecular mechanisms [22,23]. A more general, physiological approach to this drop could be the decrease of sympathetic tone due to the body posture during sleep [24]. …”
