Mechanism of surface plasmon resonance sensing by indirect competitive inhibition immunoassay using Au nanoparticle labeled antibody

Abstract: We investigated the use of a surface plasmon resonance (SPR) biosensor using an antibody (Ab) labeled with Au-nanoparticle (Ab-AuNP conjugate). As clenbuterol is a small molecule, an indirect competitive inhibition immunoassay was used. The SPR immunoassay using Ab-AuNP conjugate had an extremely low limit of the detection (LOD) with a magnitude of 0.05 ppt (0.05pgmL), which was 40 times lower than that of unlabeled Ab. To identify the key factor in determining the LOD of the indirect competitive inhibition im… Show more

“…This change in absorbance was due to the different RI of the HIgG solution compared to the buffer (DI + PBS) solution. The absorbance dynamics of HIgG follow standard biomolecular association, where exponential growth in absorbance was observed.…”

“…Wang et al [82] developed a SPR to detect ractopamine in swine urine through indirect competitive assay, with an LOD of 0.09 ng/mL. Kabiraz et al [83,84] built SPRs sensors to detect clenbuterol using Au nanoparticles to label the primary antibody against clenbuterol or the secondary antibody. The LODs were 0.05 pg/mL and 100 fg/mL, respectively, which were lower than that of unlabeled Ab.…”

“…This research group also fabricated a similar AuNPs/PDA SPR immunosensor for cardiac biomarker troponin I (cTropI) with LOD of 38 ng mL −1 , which is 67-fold lower than that of SPR biosensor only using AuNPs [176]. Kabiraz et al used AuNPs to label the antibody to develop an indirect competitive SPR immunoassay to detect the analyte with a small molecule (clenbuterol) [177]. The SPR immunoassay has an extremely low LOD (0.05 pg mL −1 ), which was 40-fold lower than that using unlabeled antibody.…”
