Mechanism of Longevity Extension of Caenorhabditis elegans Induced by Schizophyllum commune Fermented Supernatant With Added Radix Puerariae

Abstract: Schizophyllum commune (S. commune) fermented supernatant with added Radix Puerariae (SC-RP) showed significant antioxidant activity in our previous work. However, the possible lifespan and healthspan extending the capacity of Caenorhabditis elegans (C. elegans) and the underlying mechanism were not illuminated. In this study, the effect of SC-RP on extending the lifespan and improving stress resistance of C. elegans were examined. Additionally, the underlying lifespan extending molecular mechanisms of SC-RP we… Show more

“…The day of transfer was recorded as Day 0, and the locomotion of the C. elegans was quantified on the 4th, 8th, and 12th days. A sinusoidal motion was defined as a one-wavelength shift relative to the long axis of the body (23). The mobility was designated to three levels as following standards: Class A, animals move spontaneously and smoothly; Class B, animals don't move unless stimulated; Class C, animals don't move but wiggle their noses or tails in response to touch (24).…”

“…Three regular factors for different inoculum amounts (2, 5, 8, and 11%, v/v), temperature (22,25,28, and 31 • C), pH (5, 6, 7, and 8) were selected to increase the phenolic content. While one of the variables was being evaluated, the other fixed parameters were set to an inoculum size of 5% (v/v), a temperature of 25 • C, and an initial pH of 6.0.…”

“…The advantages of bidirectional fermentation are that it better promotes the extraction of active ingredients, increases the efficacy of fermentation, and can even produce new active substances ( 20 ). In our recent work, exopolysaccharide yields and antioxidant activities were significantly improved using Schizophyllum commune fermented products by bidirectional fermentation with Radix Puerariae ( 21 ), and these metabolites could extend the lifespan and health-span of Caenorhabditis elegans ( 22 ).…”

“…At the same time, SPG also exhibits certain anti-virus activities [ 18 ] and moisture properties, which can provide a continuous moist environment for wound healing because of its high water retention capacity. Additionally, the yield of SPG was significantly improved by bidirectional fermentation and the response surface methodology in our recent work [ 19 ]. The possible anti-aging activities of the fermentation supernatant have been illuminated [ 20 ].…”
