Mechanism of Curcuma wenyujin Rhizoma on Acute Blood Stasis in Rats Based on a UPLC-Q/TOF-MS Metabolomics and Network Approach

Abstract: Rhizome of Curcuma wenyujin, which is called EZhu in China, is a traditional Chinese medicine used to treat blood stasis for many years. However, the underlying mechanism of EZhu is not clear at present. In this study, plasma metabolomics combined with network pharmacology were used to elucidate the therapeutic mechanism of EZhu in blood stasis from a metabolic perspective. The results showed that 26 potential metabolite markers of acute blood stasis were screened, and the levels were all reversed to different… Show more

“…The detailed information of disordered metabolic pathways is presented in . In this study, the pathway impact value and P value were set to 0.10 and 0.01 according to previous researches [31, 37, 38]. 5 of the metabolic pathways (steroid hormone biosynthesis, linoleic acid metabolism, alpha-linolenic acid metabolism, retinol metabolism, and arachidonic acid metabolism) showed evident changes in BSS rats according to impact >0.1 and P < 0.01, which indicated they might be significantly associated with BSS's development and RWCI intervention.…”

“…Similar to the PCA score map, ND and HFD were well separated from each other, whereas HFD+PRbs and HFD+Rbs partially overlapped and were more closely related to ND ( Figure 3B). After normalizing the obtained data with the Pareto scaling method [23,24], we analyzed the data with unsupervised principal component analysis (PCA), a multivariate statistical method which reduces the number of observations through dimensionality reduction into a few comprehensive indicators while it preserves the main information of the original variables ( Figure 3A). PC 1 and 2 explain 36.2% and 19.5% of the metabolic variance of serum, respectively.…”

“…To evaluate the validity of the established OSC-PLS-DA model, 200 permutation tests and repeated 2-fold cross-validation (2CV) were performed 45,46 . The quality of the model was assessed based on R2 and Q2 scores, as R2 explained the total variation of the data, and Q2 described the predictive ability of the model 47 . Differential metabolites were assessed based on VIP values (VIP > 1) and t-tests (P < 0.05) 48 .…”
