Mechanism of action and therapeutic potential of dimethyl fumarate in ischemic stroke

Abstract: Dimethyl fumarate (DMF) is an immunomodulatory drug currently approved for the treatment of multiple sclerosis and psoriasis. Its benefits on ischemic stroke outcomes have recently come to attention. To date, only tissue plasminogen activators (tPAs) and clot retrieval methods have been approved by the FDA for the treatment of ischemic stroke. Ischemic conditions lead to inflammation through diverse mechanisms, and recanalization can worsen the state. DMF and the nuclear factor erythroid-derived 2-related fact… Show more

“…Moreover, BG-12 appears to suppress platelet function and thrombus formation (Chu et al, 2023). Several other reports suggest a potential ameliorative role for dimethyl fumarate in Alzheimer's disease, ischemic stroke, systemic lupus erythematosus, traumatic brain injury, among others (Cheng et al, 2023;Mauro et al, 2023;Owjfard et al, 2023). On the other hand, Oltipraz is currently in phase III trial for the treatment of nonalcoholic fatty liver disease (Robledinos-Antón et al, 2019) and is being proposed as potential agent for the management of Thoracic aortic aneurysm and dissection and osteoporosis (Wang et al, 2022;Yang et al, 2022).…”

“…Seemingly minor details buried in a patient’s account may shed light on occult pathologies underlying their complaints. In cases of nonspecific limb discomfort, benign-appearing symptoms may signal retained foreign bodies or other occult disease processes [ 7 9 ].…”
