Mechanical Scarification of Dodder Seeds with a Handheld Rotary Tool

Abstract: Dodder seeds are physically dormant because of hard seed coats and do not readily germinate without scarification. Reliable methods of scarification for small lots of dodder seed are needed to facilitate laboratory, greenhouse, and field research projects. Dodder seed was scarified for varying times using a handheld rotary tool at the 10,000 rpm setting with a conical grinding-stone bit attached. Percentage of germination and weight change of seeds were assessed using scarification times between 0 and 4 min at… Show more

“…Dodder seeds were scarified in batches of 100 (0.01 g) in a 2 mL microcentrifuge tube for approximately 3 minutes using a small dremel tool [ 33 ]. Seeds were then rinsed with tap water using a fine mesh strainer, placed in Petri dishes lined with 90 mm moistened filter paper, sealed with Parafilm (Bemis Company Inc., Oshkosh, WI, USA), and placed in an incubator at 23°C until seed germination approximately 2 days later.…”

“…This is triggered by the micropyle when a suitable environmental condition emerges. A sim claim is proposed by Ghantous Sandler [16][17][18] in their study on dodder seed dormancy. They argue that seed scarification of capsuled dodder berries is a must for them to germinate.…”
