Mechanical Nanosensor Based on FRET within a Thermosome: Damage‐Reporting Polymeric Materials

“…Time-domain fluorescence lifetime imaging microscopy (FLIM) for additional FRET sensor characterization was implemented with an inverted Zeiss LSM510 Axiovert 200M microscope with a Plan NeoFLUAR 40×/1.3 NA DIC oil-immersion objective lens, equipped with a TCSPC controller (SPC-830 card; Becker & Hickl, Berlin, Germany) as described previously 32 . CFP was excited with 440 nm light generated by frequency doubling of 880 nm pulses from a mode-locked Ti:sapphire laser(Mai-Tai, Spectra-Physics, 120–150 fs pulse width, 80 MHz repetition rate, and Frequency Doubler and Pulse Selector, Spectra-Physics, Model 3980).…”

“…Using two complementary hetero-bifunctional linkers confers the further advantage that cross-linking cannot occur. Such a system was used to specifically and quantitatively conjugate oligonucleotides to antibodies [63] and to bind proteins to other proteins [64] in aqueous solution. Because this conjugation reaction is carried out under mild conditions in aqueous buffer, without any additional catalyzing agents (e.g., in contrast to the copper catalyzed azide-alkyne click reaction), the resulting conjugates are applicable for pharmaceutical and therapeutic purposes.…”

“…[1][2][3][4] Mechanochromic polymeric materials, which change color under an applied force, have been well studied. [5][6][7][8][9] Spiropyran (SP) is one of the most promising mechanophores, which is colorless and undergoes a 6-π electrocyclic ring-opening reaction to form colored merocyanine (MC) under external force. 10,11 SP based mechanochromic materials can be obtained by covalent [10][11][12][13][14][15] and non-covalent bonding to the matrix.…”
