Mechanical and physio-biological properties of peptide-coated stent for re-endothelialization

Abstract: Background: The aim of this study was to characterize the mechanical and physio-biological properties of peptidecoated stent (PCS) compared to commercialized drug-eluting stents (DESs). Methods: WKYMVm (Trp-Lys-Tyr-Met-Val-D-Met), a stimulating peptide for homing endothelial colony-forming cell was specially synthesized and coated to bare metal stent (BMS) by dopamine-derived coordinated bond. Biological effects of PCS were investigated by endothelial cell proliferation assay and pre-clinical animal study. And… Show more

“…To treat these lethal diseases, the vascular stent has been commonly used to restore blood flow. First-generation bare metal stents (BMSs), consisting of cobalt-chromium (Co-Cr) and stainless steel support the lining of the blood vessels with high mechanical properties but have been frequently reported to induce in stent restenosis (ISR) and thrombosis due to foreignbody reactions [1][2][3]. Second-generation drug-eluting stents (DESs) have been developed from BMSs to overcome this problem and provide additional drug release.…”

“…Additionally, it has shown excellent mechanical properties compared to other synthetic polymers at a more affordable price [ 20 , 21 ]. It is essential to select an adequate material for use on stents because there are many requirements that must be met, including that the material should be non-cytotoxic, resorbable, flexible, radiopaque, biocompatible, non-erodible, and it should display excellent hemocompatibility, controlled drug encapsulation and release, and sufficient radial strength [ 5 , 8 , 21 , 22 ].…”

“…Peptides targeting endothelial cells have also been investigated, including REDV (Arginine-Glutamate-Aspartate-Valine) ( Hubbell et al, 1991 ). Specific peptides to EPCs have been identified such as the disulfide cyclic octa-peptide (cGRGDdvc, also known as LXW7) ( Hao et al, 2017 ), TPS (Threonine-Proline-Serine-Leucine-Glutamate-Glutamine-Arginine-Threonine-Valine-Tyrosine-Alanine-Lysine) ( Veleva et al, 2007 ), and WKYMVm (Trp-Lys-Tyr-Met-Val-D-Met) ( Bae et al, 2020a ). These peptides interact with the integrins -which are adhesion receptors on the cells - and activate them, resulting in enhanced cell adhesion and binding.…”
