Measuring Hemoglobin Oxygen Saturation During Graded Hypoxic Hypoxia in Rat Striatum

Abstract: We evaluated in vivo reflectance spectroscopy of visible light as a method to assess brain tissue hemoglobin oxygen saturation in rat striatum (SstrO2). Seven anesthetized and mechanically ventilated rats were subjected to incremental reduction in the fraction of inspired oxygen (Fio2): 0.35, 0.25, 0.15, 0.12, and 0.10, followed by a reoxygenation period (Group 1). At each episode, local changes in SstrO2 and in cerebral blood flow (LCBF) were simultaneously determined in the two striatal regions, using reflec… Show more

“…Epinephrine was required to counteract the adverse effects of both anaesthesia and hypoxic hypoxia on the cardiovascular system. 20 Sodium bicarbonate was used to prevent metabolic acidosis. For all experiments, the rats were placed in pronation; the head was secured via ear and teeth bars.…”

“…Premature babies are the most likely to suffer prolonged periods of hypoxemia. Although data from neonatal rats are sparse, studies in adult rats suggest that our model would have led to an oxygen saturation of 65–70% 38, 39. These levels are certainly well below the therapeutic O 2 saturation (i.e., between 85–95%) currently used in many neonatal intensive care units 40.…”

“…The same process was done with the reflectance data. The reflectance spectra, after normalization and correction (divided by a well-known reference spectrum), have been fitted with a combination of Hb and HbO2 absorption spectra (19), which can provide an index of oxygen saturation of the tissue (SO2) -results not shown. Moreover, the ratio between channel 1 and 2 normalized spectra can be integrated over a wavelength range (from 472nm to 540nm) leading to a quantitative parameter evolving with the FITC depth.…”
