Measuring circular dichroism in a capillary cell using the b23 synchrotron radiation CD beamline at diamond light source

Abstract: Synchrotron radiation circular dichroism (SRCD) is a well-established method in structural biology. The first UV-VIS beamline dedicated to circular dichroism at Diamond Light Source, a third generation synchrotron facility in South Oxfordshire, has recently become operational and it is now available for the user community. Herein we present an important application of SRCD: the CD measurement of protein solutions in fused silica rectangular capillary cells. This was achieved without the use of any lens between… Show more

“…CD experiments were performed using both the nitrogen-flushed Module B end-station spectrophotometer at B23 Synchrotron Radiation Circular Dichroism (SRCD) Beamline at the Diamond Light Source424344 and a JASCO J-715 spectropolarimeter (Jasco, MD, USA). For SRCD measurements, the M2AH peptide or a scrambled control peptide was diluted in water to a final concentration of 500 μM.…”

“…SRCD experiments were performed using a nitrogen-flushed Module B end-station spectrophotometer at B23 Synchrotron Radiation CD Beamline at the Diamond Light Source (35,36). McjD samples at 0.1 mg/ml were prepared in 20 mM Tris, pH 7.5, 8 mM NaCl containing 0.03% DDM or lipids PE, PG, or PL.…”

“…One of the unique feature of Diamond B23 beamline for SRCD is the highly collimated light beam (about 0.5 mm (H) × 0.3 mm (V) FWHM), which has been exploited for measurement of samples with limited availability using small aperture cells of low volume capacity (from 40 μl for 1 cm to 800 μl for 10 cm pathlengths)111213. This feature also enabled the measurement to be conducted on the centre of each well of a multi-plate without any significant distortion effect produced by the meniscus of the liquid (Fig.…”
