MEASUREMENTS OF PHYTOPLANKTON‐PROTEIN CONTENT WITH THE HEATED BIURET‐FOL1N ASSAY1,2

Abstract: The heated biuret‐Folin method for determining protein consistently measures 90% of the total nitrogen of filtered algae samples as protein‐N without the need of mechanical disruption as long as the heating period in biuret is 100 min at 100 C. Data indicate this protein assay measures total protein on all species tried and for naturally occurring mixtures of species plus detritus. Dilute algal suspensions with as little as 0.05 μg‐atom particulate protein N.liter ‐1 can he concentrated by fltration on glass f… Show more

“…A time-course experiment was conducted during which triplicate samples from each culture were taken morning and evening (just before lights-on and lights-off) for analysis of chlorophyll, nitrate, protein, and carbohydrate. Chlorophyll and nitrate were analyzed as described for tank experiments, protein was analyzed by the Biuret-Folin method of Dorsey et al (1978) using bovine serum albumin (BSA) as standard, and carbohydrate was measured as glucose equivalents by the phenolsulphuric acid method of Dubois et al (1956). Samples were taken before and after depletion of nitrate from th.e culture medium.…”

“…Protein concentration was determined according to Strain et al (2006) based on Dorsey et al (1978) modified from Lowry et al (1951). Briefly, 1 mg freeze-dried phytoplankton was hydrolyzed in 10 ml Biuret reagent (diluted 1:1 with distilled water) at 100°C for 1 h and then mixed with 0.5 ml Folin-phenol reagent.…”
