Measurement of superoxide dismutase, catalase and glutathione peroxidase in cultured cells and tissue

Abstract: Cells contain a large number of antioxidants to prevent or repair the damage caused by ROS, as well as to regulate redox-sensitive signaling pathways General protocols are described to measure the antioxidant enzyme activity of superoxide dismutase (SOD), catalase, and glutathione peroxidase. The SODs convert superoxide radical into hydrogen peroxide and molecular oxygen, while the catalase and peroxidases convert hydrogen peroxide into water. In this way, two toxic species, superoxide radical and hydrogen per… Show more

“…SOD enzyme converts superoxide radicals into molecular oxygen and hydrogen peroxide, while CAT converts hydrogen peroxide into water and oxygen (Firuzi et al, 2011;Weydert and Cullen, 2010). In the present study, the liver of both SOD and CAT enzyme activities were significantly lower in the APAP-only treated group.…”

“…The levels and enzyme activities of Mn-and CuZn-superoxide dismutases (SOD1 and SOD2) were determined by Western blot analysis and by in-gel activity assay as described previously (68). To determine steady-state levels of these enzymes by Western blotting, we used an anti-SOD1 antibody from Calbiochem and an anti-SOD2 antibody from Upstate.…”

“…SOD converts superoxide radicals into hydrogen peroxide and molecular oxygen (O 2 ), while the CAT and POD convert hydrogen peroxide into water, and in the case of CAT, to oxygen and water [45]. Capsaicinoids are products of the phenylpropanoid and fatty acid synthesis pathways [46].…”
