Measurement of gp210 autoantibodies in sera of patients with primary biliary cirrhosis

Abstract: Primary biliary cirrhosis (PBC) is an autoimmune liver disease with unknown etiology. Patients with PBC have antimitochondrial autoantibodies (AMA) and additionally 50% of them have antinuclear antibodies (ANA). A 15-amino acid fragment (DRKASPPSGLWSPAY) from the C-terminal part of the nuclear envelope glycoprotein gp210 has been proposed as one of the antigenic targets for ANA. The aim of this work was to develop an immunoenzymatic assay for determination of gp210 autoantibodies using for its binding a synthe… Show more

“…A total of 25 studies on GP210 (19 studies in English [4] , [5] , [9] , [10] , [12] , [13] , [18] [21] , [26] , [28] [35] and six in Chinese [6] , [14] , [15] , [17] , [22] , [23] ) were published between 1988 and 2013 ( Table 1 ), and involved a total of 2,806 PBC patients and 7,494 controls. Of these 25 studies, 15 were carried out in Asia [5] , [6] , [9] , [12] – [15] , [17] , [19] , [20] , [22] , [23] , [29] , [31] , [33] , five in Europe [10] , [28] , [30] , [32] , [35] , and five in South America [4] , [18] , [21] , [26] , [34] ; 56% (14 of 25) using immunoblotting and 48% (12 of 25) with the ELISA method (one article [26] was conducted using two methods). A total of 21 studies on SP100 (13 in English [4] , [8] , [9] , [10] , [12] , [18] …”

“…The ANA pattern in PBC includes multiple nuclear dots (MND-ANA) and nuclear membrane-associated ANAs (NM-ANAs). About 30% of patients manifest anti-nuclear envelope (NE) positivity [ 13 , 14 , 15 , 16 ]. In the majority of cases, anti-NM antibodies target gp210 and p62 proteins.…”

“…The nuclear rim pattern is indicative of antibodies directed against components of the nuclear envelope and nuclear pore complex, with gp210 being the primary target antigen in PBC [38]. Gp210 antibodies were found in approximately 25% of patients with PBC, had a specificity of greater than 95% [36,39] and were associated with an increased risk of hepatic failure [10,40,41] and a higher Mayo risk score [42]. In the present study, 27% of our PBC cohort reacted with gp210, but there was no significant association with a higher Mayo risk score or liver decompensation.…”
