Our interest in providing parameters for the development of quantitative structure physiologically based pharmacokinetic/pharmacodynamic (QSPBPK/PD) models for assessing health risks to carbamates (USEPA 2005) comes from earlier work with organophosphorus (OP) insecticides (Knaak et al. 2004). Parameters specific to each carbamate are needed in the construction of PBPK/PD models along with their metabolic pathways. Parameters may be obtained by (1) development of QSAR models, (2) collecting pharmacokinetic data, and (3) determining pharmacokinetic parameters by fitting to experimental data. The biological parameters are given in Table 1 (Blancato et al. 2000). An improved version of the disposable multichannel immunochemical biosensor for the herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) based on a screen-printed amperometric transducer and monoclonal antibodies (MAb) against 2,4-D is reported. Entrapment within a thin Nafion film was used for the direct immobilization of MAb at the electrode surface. The amount of the tracer (2,4-D conjugated to acetylcholinesterase) bound in a competitive immunochemical reaction was determined amperometrically using acetylthiocholine iodide as substrate. The measuring procedure (times of incubation with tracer and substrate, pH, tracer concentration) was optimized. The sensor was able to detect less than 0.01 pgL of free 2,4-D in water. One analysis (8 samples) was completed in 30 min (20min for immunochemical reaction, 5 min incubation with substrate, 5 min measurement). The performance of the immunosensor (two configurations) was evaluated on real samples (tap water) with added 2,4-D. The determined amounts (mean values 0.097 to 0.105 and 0.89 to 1.13) corresponded well with the added contents of 2,4-D (0.100 and 1.00 pgL, respectively).

Detection of carbamate pesticides in vegetable samples using cholinesterase‐based biosensors

Biosensors for determination of carbamates in vegetables based on five different cholinesterases as biorecognition elements and a screen-printed electrode system as an amperometric transducer were developed. Measurements were simply performed by dropping solutions (either sample or substrate) directly on the biosensor. The response of biosensors to selected carbamates (aldicarb, carbaryl, carbofuran, methomyl and propoxur) was characterized. The performance was evaluated on extracts of potatoes and carrots, the results from the AChE biosensor and a standard HPLC procedure were compared. Finally, the biosensor was used for the direct analysis of vegetable juices without any pretreatment steps. In this case, 10lg/L levels of added carbofuran and propoxur were reliably identified. The whole procedure takes less than 20min including 10 min incubation with samples. The concentrations of carbamates determined with biosensor agreed well for carbofuran. Lower response was observed for propoxur.
