Measles virus antigens induce both type-specific and canine distemper virus cross-reactive cytotoxic T lymphocytes in mice: localization of a common Ld-restricted nucleoprotein epitope

Abstract: We have studied the induction of the cytotoxic T lymphocyte (CTL) response to measles virus (MV) antigens expressed as vaccinia virus (VV) recombinants in a murine model. In C3H mice (H-2 k) only the nucleoprotein (NP) induced a CTL response and this was shown to be cross-reactive with the closely related canine distemper virus (CDV). The presentation of this antigen was shown to be Kk-restricted. In BALB/c mice (H-2d), both the haemagglutinin (HA) and the NP induced a strong CTL response, the former being ser… Show more

“…66 following challenge, but this was only demonstrated in cells expressing all virus antigens, suggesting that this CTL activity was directed against another RPV antigen. Thus far, the role of CTLs in controlling morbillivirus infection has only been examined in MV-induced encephalitis models in rodents in which virus growth is limited to the central nervous system (Beauverger et al, 1993 ;Niewiesk et al, 1993). Other authors have questioned the role of CTLs in protection against MV-induced encephalitis in mice and emphasized the importance of CD4 + T-cells (Finke & Liebert, 1994).…”

“…The approach to the evaluation of the role of structural proteins in the induction of protective immunity in measles has obvious implications for the rational design of new generations of vaccine and is being actively pursued in murine models (13)(14)(15)(16)(17)(18). Although information is beginning to emerge from studies in humans (19)(20)(21), an extension of the research on immune responses to measles gene products during the disease in genetically different human populations in endemic areas will provide information for the development of new vaccines and for understanding the pathogenesis of the infection and the basis of vaccine failure.…”
