MDCK (Madin-Darby Canine Kidney) Cells: A Tool for Membrane Permeability Screening

Abstract: The goal of this work was to investigate the use of MDCK (Madin-Darby canine kidney) cells as a possible tool for assessing the membrane permeability properties of early drug discovery compounds. Apparent permeability (Papp) values of 55 compounds with known human absorption values were determined using MDCK cell monolayers. For comparison, Papp values of the same compounds were also determined using Caco-2 cells, a well-characterized in vitro model of intestinal drug absorption. Monolayers were grown on 0. 4-… Show more

“…(27). In contrast, the collecting duct-derived MadinDarby canine kidney (MDCK) cell line is routinely used for drug permeability and transport studies (following transfection with specific human drug transporters), typically in the context of oral drug absorption and brain penetration (28). Similarly, Caco-2 cells are widely used as an in vitro model of intestinal, as well as blood-brain barrier permeability, and have recently been proposed as a possible in vitro model of passive renal tubular reabsorption (29).…”

“…represents the flux across the monolayer (permeability rate), A (cm 2 ) the surface area of the monolayer, V (cm 3 ) the volume of the receiver chamber, and C o (μmol/L) the initial concentration in the donor compartment (25). Results are presented as mean P app ± SD (n = 3).…”

“…MDCK cells and Caco-2 cells have been reported to share many common epithelial cell characteristics (Putnam et al, 2002). A good correlation between the permeation of passively absorbed drugs across Caco-2 cells and MDCK cells suggests that MDCK model may be useful in place of Caco-2 cells as a monolayer model for intestinal mucosa (Irvine et al, 1999). In the late 1990s, a MDCKII-MDR1 cell line was generated by transfecting human mdr1 gene into MDCK cells.…”
