MCM3AP Is Transcribed from a Promoter within an Intron of the Overlapping Gene for GANP

Abstract: MCM3 acetylase (MCM3AP) and germinal-centre associated nuclear protein (GANP) are transcribed from the same locus and are therefore confused in databases because the MCM3 acetylase DNA sequence is contained entirely within the much larger GANP sequence and the entire MCM3AP sequence is identical to the carboxy terminus of GANP. Thus, the MCM3AP and GANP genes are read in the same reading frame and MCM3AP is an N-terminally truncated region of GANP. However, we show here that MCM3AP and GANP are different prote… Show more

“…S7). However, since full‐length GANP(P443S) protein was not detectable (data not shown), it is conceivable that caspase‐mediated cleavage raised the level of HR‐stimulatory C‐terminal fragments mimicking MCM3AP [Wickramasinghe et al., ]. Altogether, elevated full‐length GANP/MCM3AP ratios in patient cells and WT‐GANP expressing parental cells represent a possible explanation for the observed HR decreases.…”

“…Strangely, MCM3AP is transcribed from a promoter within an intron of GANP and has an unrelated function within the nucleus 104 . These features are consistent with the spatial distribution of GANP, which is concentrated at the nuclear envelope but is also present throughout the nucleus 12,56,105 .…”

“…A shorter isoform of GANP, GANP/MCM3AP (ref. 37), is likely to encode an 80-kDa protein associated with MCM3 of the DNA helicase MCM-complex composed of MCM2-MCM7 (refs 28, 38) and possesses HAT activity. Based on the Sac3-homology domain, GANP has been shown to play important roles in the mRNA export of mammalian cells3539.…”
