MBD2 regulates differentiation and function of Th17 cells in neutrophils- dominant asthma via HIF-1α

Abstract: BackgroundT helper 17 (Th17) cells have proven to be crucial in the pathogenesis of neutrophils-dominant asthma. Hypoxia inducible factor-1α (HIF-1α) is involved in allergic responses in asthma. Our previous studies indicated that Methtyl-CpG binding domain protein 2 (MBD2) expression was increased in asthma patients. The aim of the present study is to understand how MBD2 interacts with HIF-1α to regulate Th17 cell differentiation and IL-17 expression in neutrophils-dominant asthma.MethodsA neutrophils-dominan… Show more

“…Moreover, HIF-1α has been reported to facilitate the differentiation of Th17 cells [27], and may serve as an important signaling molecule for the induction of asthma through Th17 cells [14,15]. HIF-1α can also cause asthma by means of airway smooth muscle remodeling [28,29], which is in line with our ndings.…”

“…Other studies have illustrated that deficiency of HIF-1α can reduce eosinophil infiltration, goblet cell hyperplasia, and levels of cytokines IL-4, IL-5, and IL-13 in the lungs of OVA-induced asthma models [ 13 ], further highlighting the importance of the elevated levels of HIF-1α in asthma. Moreover, HIF-1α has been reported to facilitate the differentiation of Th17 cells [ 30 ], and may serve as an important signaling molecule for the induction of asthma by promoting Th17 cells [ 16 , 17 ]. HIF-1α can also cause asthma by means of airway smooth muscle remodeling [ 31 , 32 ], which is in line with our findings.…”

“…CpGs cg11316510 and cg09573852 on genes RARA (retinoic acid receptor alpha) and IKBKB (Inhibitor of Nuclear Factor Kappa B Kinase), respectively, were among the IOW-ALSPAC consistent CpGs but not on the list of IOW-BAMSE consistent CpGs. Their significant involvement in lung function, as well as lung function development and pulmonary diseases such as asthma and COPD indicated the potential importance of these two CpGs and their mapped genes [11,[61][62][63][64][65][66][67][68][69][70][71]. RARA is the predominant isotype of the retinoic acid receptor (RAR) identified in alveolar type II epithelial cells and components of the retinoic acid signaling pathway [63][64][65][66][67][68].…”
