Mature Let-7 miRNAs fine tune expression of LIN28B in pluripotent human embryonic stem cells

Abstract: MicroRNAs (miRNA) are central regulators of diverse biological processes and are important in the regulation of stem cell self-renewal. One of the widely studied miRNA-protein regulators is the Lin28-Let-7 pair. In this study, we demonstrate that contrary to the well-established models of mouse ES cells (mESC) and transformed human cancer cells, the pluripotent state of human ES cells (hESC) involves expression of mature Let-7 family miRNAs with concurrent expression of all LIN28 proteins. We show that mature … Show more

“…The mechanistic explanation for this is not known; however, recent studies have demonstrated that, in some CRC cell lines, overexpression of pluripotency factors (OCT3/4, SOX2, and KLF4) can lead to tumors with increased differentiation (Oshima et al 2014). This is in line with our finding, since LIN28B is also a pluripotency factor (Rahkonen et al 2016;Zhang et al 2016;Tsanov et al 2017), although this does not mean that pluripotency and differentiation are necessarily linked in this context.…”

“…LIN28B is a critical pluripotency factor and is engaged in diverse cancer types, including CRC (Rahkonen et al 2016;Zhang et al 2016;Tsanov et al 2017). LIN28B acts to a major extent through the post-transcriptional down-regulation of the let-7 miR family.…”

“…As mentioned above, miRNAs function at the post-transcriptional level and have been demonstrated to be a promising approach for fine-tuning the excessive expression of their target genes in several biological processes (Rahkonen et al, 2016 ; Roy, 2016 ; Su et al, 2016 ). In addition, intranasal delivery of miRNA mimics and inhibitors, which can bypass the blood-brain barrier to deliver drugs into the brain, has been used recently to treat several neurological diseases (Ma et al, 2016 ; Lee et al, 2017 ; Tao et al, 2017 ).…”

“…Можно предположить, что эти миРНК задействованы в процессе поддержания плюрипотентности у крысы. Кроме того, в данной сети участвуют миРНК, вовлеченные в процесс негативной регуляции плюрипотентного состояния клеток мыши и человека, такие как, например, miR145 и представители семейства let7 (Xu et al, 2009;Rahkonen et al, 2016). В сети представлены также миРНК семейства miR181, регулирующие Tcl1a и Wnt3a.…”
