Maturation of the Epithelial Na+ Channel Involves Proteolytic Processing of the α- and γ-Subunits

Abstract: The epithelial Na ؉ channel (ENaC) is a tetramer of two ␣-, one ␤-, and one ␥-subunit, but little is known about its assembly and processing. Because co-expression of mouse ENaC subunits with three different carboxyl-terminal epitope tags produced an amiloride-sensitive sodium current in oocytes, these tagged subunits were expressed in both Chinese hamster ovary or Madin-Darby canine kidney type 1 epithelial cells for further study. When expressed alone ␣-(95 kDa), ␤-(96 kDa), and ␥-subunits (93 kDa) each prod… Show more

“…Recently, new evidence has emerged that suggests that activation of ENaC involves direct furin-mediated cleavage of ␣-and ␥-ENaC subunits within the biosynthetic pathway (13,14). These results suggest that changes in biochemical levels of full-length ENaC subunits may not correlate directly with modulation of sodium currents.…”

“…The generation of cDNAs for mouse ENaC (mENaC) subunits epitope-tagged on their carboxyl termini (V5 tag on ␣-and ␥-mENaC; FLAG tag on ␤-mENaC) is described in Ref. 13. The constructs were amplified by PCR and subcloned downstream of the tetracycline operon in the pAdtet vector using HindIII and SpeI restriction sites.…”

“…Cleavage by Subtilisin-To further examine ␣ ENaC processing, we mutated the terminal arginine (P1) in both identified serine protease (furin) cleavage sites (Arg-178 and 204) to alanines (2). This ␣ENaC construct is referred to as ␣FM.…”

“…2 Activation occurs by direct mechanisms that induce channel subunit cleavage (1, 2) as well as those that are cleavage-independent but may involve cleavage of protease-activated membrane receptors (3). Channel cleavage studies have established that cellular proteases such as furin endogenously cleave the channel ␣ and ␥ subunits.…”

“…The ENaC is also a target for a variety of post-translational modifications, including proteolytic cleavage in its ECD, which increases its activity (40). Similar to this activating mechanism, it is possible that intracellular proteases may cleave off the N-terminal part of BASIC upon maturation or application of a certain stimulus and thus lead to a more active channel compared with resting conditions.…”
