Matrix metalloproteinase gene polymorphisms and oral cancer

Abstract: Since oral squamous cell carcinoma (OSCC) is the most prevalent malignant cancer in the oral cavity, several researches have been performed to study the role of important enzymes in this disease. Among them, the matrix metalloproteinases (MMPs) are highlighted, due to the fact that they are proteinases responsible to degrade many extra-cellular matrix components, making possible the invasion of neoplasic cells. Important tools in cancer prognosis have been utilized aiming to correlate high levels of MMPs and O… Show more

“…61,62 In another study, by Chaudhary et al, the authors detected a higher frequency of MMP1-promoter genotypes with the 2G allele, which is associated with higher enzymatic activity, in patients with HNSCC or the preneoplastic condition, oral submucous fibrosis. 63 In addition, elevated expression of the 5A allele of the MMP3 gene has been associated with an increased risk of oral SCC development.…”

“…MMP1 encodes an enzyme that plays an active role in the breakdown of interstitial collagen, including types I, II, and III, and is secreted by a variety of cell types such as mesenchymal cells (either VSMCs or fibroblasts), macrophages and epithelial cells . MMP1 is also important for the cell‐matrix interactions, and its rs1799750 polymorphism characterized by deletion/insertion of a guanine in the promoter seems to affect transcriptional activity and expression level . However, our meta‐analysis indicated no obvious association between MMP1rs1799750 polymorphism and AAA risk.…”

“…These polymorphisms may affect the production or gene expression of these MMPs in an allele-specific manner (Piccoli et al, 2007;Tsuchiya et al, 2009;Wieczorek et al, 2013;Yaykas xli et al, 2014). Furthermore, there is an increasing evidence indicating that these functional polymorphisms may contribute to interindividual differences in a wide spectrum of cancer susceptibility (Decock et al, 2008;Pereira et al, 2012;Wieczorek et al, 2012).…”
