BackgroundBladder cancer (BC) is an aggressive disease with a poor prognosis. A bladder tumor, like other malignant neoplasms, is characterized by the presence of both cancer cells and stromal cells which secrete cytokines, chemokines, growth factors, and proteolytic enzymes. One such class of proteolytic enzymes are serine proteases, which take part in the tumor microenvironment formation via supporting and contributing to tumor progression. This study aims to evaluate the proteolytic activity and serine protease contribution in plasma from BC patients.MethodsThe research involved patients of Alexandrovsky city clinical hospital aged 52–76 with transitional cell carcinoma of the bladder. All examined patients were divided into five groups: the control group included conditionally healthy donors, while other patients were grouped according to their tumor stage (I, II, III and IV). Plasma plasminogen levels were determined by enzyme-linked immunosorbent assay, and the potential activity was measured by chromogenic plasminogen assay. Serine proteases fractions were obtained by the affinity chromatography method, and enzyme concentration in the selected fractions were determined by the Bradford method. Serine proteases distribution was investigated by electrophoresis in a polyacrylamide gel.ResultsIt was determined that the concentration, potential activity of plasminogen, and the total amount of serine proteases in plasma from BC patients were greater than the values of the corresponding indicators in healthy donors. This could be one of the factors contributing to increased proteolysis seen in the process of carcinogenesis. Plasminogen concentration in BC patients with stage IV disease; however, displayed a tendency to be reduced compared to earlier stages, and the potential activity of plasminogen was significantly lower in patients with stages III – IV BC. Futhermore, a tumor stage specific gradual decline in the serine protease plasma content was shown. The results of electrophoretic analysis established a significant diminishment in the percentage of high molecular weight components (under non-reducing conditions) and their complete disappearance (under reducing conditions) in plasma serine protease fractions from BC patients. A decline in the percentage of heavy and light plasmin chains in BC patients was also observed. Additionally, a rise in the degraded forms of plasminogen/plasmin content was seen in BC samples, as well as the presence of fractions corresponding to trypsin and NE (under non-reducing conditions) that were absent in the control samples.ConclusionThe results indicate significant changes in the proteolytic activity of plasma, from BC patients when compared to healthy controls, which is accompanied by alterations in serine protease distribution caused by tumor microenvironment pecularlities at the different stages of oncopathology.

Among the entire public of fractures of long bones, more complications of reparative osteogenesis occur fragmental fractures. Their significant defects, which arise as a result of the removal of non-viable fragments, cause disturbances in local hemodynamics and microcirculation and loss of osteoconduction in the area of injury, lead to a loss of the reparative potential of bone tissue in the defect zone, and the methods of osteosynthesis do not provide restoration of osteoconduction and do not have osteointegration properties. For osteosis, a number of composite materials have been proposed, among which ceramics based on calcium phosphates are considered the most promising, which is confirmed by previous clinical, radiological and histomorphological studies. The aim of the study is to determine the dynamics of the blood levels of TGF-β and interleukins IL-1β and IL-10 for osteosis replacement of splinter bone fractures in dogs with calcium phosphate ceramics doped with silicon. Material and research methods: a control (n = 7) and research group (n = 7) dogs with accidental splinter fractures of long bones were formed. In both groups, extracortical osteosynthesis was performed with a support plate from an unalloyed titanium alloy. In the control group, bone defects were left to heal under a blood clot, and in the experimental group they were replaced with ceramics (GTlKg-3). Blood samples were taken after trauma no later than on the 48th day, and on the third, twelfth, twenty-first, forty-second and 60th days after surgical treatment. The enzyme immunoassay was used to determine the serum levels of interleukins IL-1β and IL-10 and transforming growth factor (TGF-β) тhat for a bureta reaction is a bit of a home-made bottle. The study results showed an anti-inflammatory cytokine profile with physiological norms in dogs. In case of bone injury, it becomes pro-inflammatory in nature, triggers the process of osteoresorption in the fracture area. At the same time, during reparative osteogenesis after extracortical osteosynthesis for splinter fractures, proinflammatory cytokinemia acquires a permanent character with a peak on the forty-second day, and this implies inhibition of metaplasia of cartilaginous regenerate into bone, that is, it slows down the consolidation of the fracture. However, in the case of osteosis with ceramics, the level of pro-inflammatory cytokinemia is significantly lower and continues during the first 12 days with a peak on the third day, during the inflammatory-resorptive stage. However, the key in understanding the effectiveness of osteosis was the dynamics of TGF-β, which showed its biphasic hyperreactivity in control animals, while in experimental animals, its balanced production was noted. Conclusion. Osteosis replacement of bone defects in long tubular bones in dogs with hydroxyapatite with β-tricalcium phosphate ceramics doped with silicon is accompanied by moderate cytokinemia in accordance with the course of the inflammatory-resorptive stage and the phase of remodeling with a peak of induction of transforming growth factor during the period of active osteogenesis forty-second course of reparative osteogenesis.

The content of the plasminogen activation system components (plasminogen, plasminogen tissue activator, and its inhibitor PAI-1) was investigated as well as the concentration of thrombomodulin, protein C, matrix metalproteinases (MMP -1, -2, -3, -8, -9, -10) and their inhibitor TIMP, growth factors content (such as transforming growth factor-β1, insulin-like growth factor-1, fibroblasts growth factor-2) and cytokine profile (interleukines IL-1β, -4, -6, -8, -10, tumor necrosis factor, interferon-γ in patients with chronic pancreatitis (CP) and pancreatic cancer (PC) were established. Khmelnitsky Regional Clinical Hospital patients aged 28-89 were selected for this study: 20 people with chronic pancreatitis (group CP); 20 people with pancreatic cancer (group PC); 20 conditionally healthy persons (control). Blood plasma samples and pancreatic tissue homogenates were obtained from all the patients. The studied indicators’ content in the experimental materials was determined by the enzyme-linked immunosorbent assay using appropriate antibodies. At the level of systemic circulation, a statistically significant increase was found in most of the studied parameters under the CP conditions. However, PC was characterized by an increase of two blood plasma indications only (thrombomodulin and protein C). In contrast, the pancreatic sample tissue examination for both aforementioned pathologies revealed significant changes in the content of most metalloproteinases and cytokines under PC. This indicates the development of metastasis, angiogenesis, and immunomodulation-aimed local biochemical processes in the affected organ.
