Matrix Metalloproteinase-2 Production and Its Binding to the Matrix Are Increased in Abdominal Aortic Aneurysms

Abstract: Abstract-Degradation of the elastic media is a hallmark of abdominal aortic aneurysms (AAAs). We examined the expression of 2 elastolytic matrix metalloproteinases (MMPs), MMP-2 and MMP-9, in AAA aortic tissues compared with those from atherosclerotic occlusive disease (AOD) and nondiseased control tissues. Quantitative competitive reverse transcription-polymerase chain reaction and gelatin zymography showed increased MMP-9 mRNA and protein in both AAA and AOD tissues compared with those in control tissue, but… Show more

“…A growing number of reports indicate that proteolytic breakdown of extracellular matrix by MMPs contribute to VSMCs migration 31. Moreover, mice deficient in MMP‐2 are actually resistant to aneurysm formation, indicating that MMP‐2 is a causative factor in the pathogenesis of AAA 17, 32. In line with this concept, in our study, we have seen significantly increased MMP‐2 levels in aneurysmatic tissue from our human and murine model of PPE‐induced AAA.…”

“…MMP-2 expression is elevated in human AAA (9,16,17). It is primarily produced by resident mesenchymal cells in AAA (9).…”

“…Increased proteolytic activity of MMPs results in weakening of the structural matrix of the abdominal aortic wall. The MMPs that have been implicated directly in AAA are MMP-9, MMP-2, MMP-1, and MMP-12 (7)(8)(9)(10)(11)(12).…”

“…Increased activities of MMP-2, -3, -9, and -12 in AAA have also been reported. [2][3][4][5] Atherosclerotic aortic lesions from high-cholesterol diet-fed apoE-KO mice show fragmentation of the elastic lamellae and rupture of the media resulting in pseudomicroaneurysm formation. These pathological changes are not observed in mice deficient in both apoE and uPA, 6 suggesting that uPA may play a key role in matrix destruction and aneurysm formation.…”
