Aflatoxin B1 (AFB1) is a mycotoxin which causes toxicity through oxidative damage. Selenium (Se), an antioxidative agent, can antagonize some toxicity induced by oxidative stress. The aim of this work was to investigate the toxicity of AFB1 and the protective effects of Se on duckling liver in vivo. The study consisted of three groups: AFB1, AFB1Tx, and a control group. AFB1 group was administered aflatoxin intragastrically (0.1 mg/kg body weight). AFB1Tx group was administered AFB1 intragastrically (0.1 mg/kg body weight) plus sodium selenite (1 mg/kg body weight). The control group was given the same volume of dimethyl sulfoxide (DMSO) via intragastric intubation. All three groups received daily administrations for 28 days. Blood samples were obtained on the 14th, 21st, and 28th days of post-administration, and the serum alanine aminotransferase (ALT) and aspartate transaminase (AST) were evaluated. A high level of serum ALT and AST was observed in AFB1 group. The activity of ALT and AST was significantly lower in Se treatment group than those in AFB1 group. Liver samples were collected on the 14th, 21st, and 28th days of post-administration, and concentrations of Bcl-2, Bax, caspase-3, and p53 were measured. Increased expression level of Bax, caspase-3, and p53 and decreased Bcl-2 expression level and Bcl-2/Bax ratio were observed in AFB1 group. Se diminished hepatic dysfunction, or damage and modulated the expression of apoptotic related proteins, in a time-dependent manner. In conclusion, concurrent treatment with Se reduced the AFB1-induced hepatic dysfunction and apoptosis.

Aim: Matrine is an alkaloid from Sophora alopecuroides L, which has shown a variety of pharmacological activities and potential therapeutic value in cardiovascular diseases. In this study we examined the protective effects of matrine against diabetic cardiomyopathy (DCM) in rats. Methods: Male SD rats were injected with streptozotocin (STZ) to induce DCM. One group of DCM rats was pretreated with matrine (200 mg·kg -1 ·d -1, po) for 10 consecutive days before STZ injection. Left ventricular function was evaluated using invasive hemodynamic examination, and myocardiac apoptosis was assessed. Primary rat myocytes were used for in vitro experiments. Intracellular ROS generation, MDA content and GPx activity were determined. Real-time PCR and Western blotting were performed to detect the expression of relevant mRNAs and proteins. Results: DCM rats exhibited abnormally elevated non-fasting blood glucose levels at 4 weeks after STZ injection, and LV function impairment at 16 weeks. The cardiac tissues of DCM rats showed markedly increased apoptosis, excessive ROS production, and activation of TLR-4/MyD-88/caspase-8/caspase-3 signaling. Pretreatment with matrine significantly decreased non-fasting blood glucose levels and improved LV function in DCM rats, which were associated with reducing apoptosis and ROS production, and suppressing TLR-4/MyD-88/caspase-8/caspase-3 signaling in cardiac tissues. Incubation in a high-glucose medium induced oxidative stress and activation of TLR-4/MyD-88 signaling in cultured myocytes in vitro, which were significantly attenuated by pretreatment with N-acetylcysteine. Conclusion: Excessive ROS production in DCM activates the TLR-4/MyD-88 signaling, resulting in cardiomyocyte apoptosis, whereas pretreatment with matrine improves cardiac function via suppressing ROS/TLR-4 signaling pathway.

This study investigated the extraction of matrine from Vietnamese sophora root and its in vitro and in vivo inhibitory effects on oral squamous cell carcinoma (OSCC) cells. Matrine was extracted from Vietnamese sophora root using microwave-assisted extraction technology and purified using macroporous adsorption resin. Human OSCC Tca8113 cells were treated with matrine with concentration of 0, 0.5, 1, 2, 4 and 8 g/L. The proliferation and apoptosis of cells were detected. OSCC cells were transplanted in the nude mice, and then the nude mice were treated with normal saline, cyclophosphamide (CTX) (20 mg·kg -1 ) and matrine (100 mg·kg -1 ), respectively. The optimized extraction parameters of matrine were obtained. The in vitro experiments showed that, matrine inhibit the proliferation of Tca8113 cells and induced the cell apoptosis. The in vivo experiments found that, the tumor inhibition rate of matrine was basically equivalent to that of CTX. On addition, compared with CTX, matrine had no obvious toxic side effect on the liver and kidney function of nude mice. These results indicate that, matrine has obvious in vitro and in vivo inhibitory effects on OSCC cells, with no obvious toxic side effect on the liver and kidney function.Keywords: extraction; matrine; oral squamous cell carcinoma. Practical Application: Matrine can be extracted from Vietnamese sophora root, and may be used to treat oral squamous cell carcinoma.
