Objective: An important predictor for infant survival is birth weight. Normal fetal growth is related to various intrauterine factors. Low birth weight is thought to have relation with oxidative stress which plays an important role in reducing the birth weight. Among the paraoxonase family PON1 protects LDL and HDL from the lipid peroxidation. This is HDL associated enzyme having antioxidant property. We aimed to evaluate the arylesterase and lactonase activity of PON1 in cord blood in relation to birth weight. We hypothesized that cord blood PON1 arylesterase and lactonase activities will be compromised in neonates having low birth weight. Methods:We included 80 neonates born in our hospital irrespective of mode of delivery as 40 cases and 40 controls. PON1 arylesterase and lactonase activity were measured using spectrophotometer.Results: Serum arylesterase activity decreased significantly in low birth weight babies (p<0.05). Linear regression analysis (R=0.595) indicates significant correlation between arylesterase and birth weight. Serum lactonase activity of PON1 also gets reduced in low birth weight babies. Its linear regression analysis showed (R=0.716) suggesting significant correlation between lactonase and birth weight. Conclusion: Reduced PON1 activity can be explained on the basis of ER stress and atherogenic changes in the placental circulation. Ours is the first study in cord blood paraoxonase activities in relation to birth weight. As the sample in our study is cord blood, it is essentially a noninvasive one. Further studies are needed in this direction to assess the effect of the oxidative stress on fetus through cord blood in its long term prospective. Objective: To investigate PON 1/Aryl activities in basketball players with or without a-T supplementation pre-and post-training. Vitamin E (a-tocopherol, a-T) reduces lipid peroxidation. Paraoxonase 1/arylesterase (PON 1/Aryl) activities are closely related to oxidation and atherogenesis. Subject/Methods: Blood was obtained from 10 players pre-(group A), post-exercise (group B) and after 1 month on a-T (200 mg per 24 h orally) supplementation pre-(group C) and post-exercise (group D). Lactate, pyruvate, muscle enzyme activities, creatine kinase, lactate dehydrogenase and total antioxidant status (TAS) were measured with commercial kits. Catecholamines and a-T were determined with high-performance liquid chromatography methods and PON 1/Aryl activities spectrophotometrically. Results: Lactate, pyruvate, muscle enzyme activities and catecholamines were increased (Po0.001) in all groups post-training. Alpha-T levels remained unaltered pre-vs post-exercise. TAS was decreased in all the groups post training. PON 1/Aryl activities were significantly decreased post-exercise (group B) (PON1: 65712 U min À1 ml À1 , Aryl: 58714 KU min À1 ml À1 ) as compared to those pre-exercise (group A) (PON1: 142716 U min À1 ml À1 , Aryl: 114712 KU min À1 ml À1 , Po0.001). In contrast, the studied enzyme activities remained practically unaltered after a-T supplementation pre-vs post-training. Both enzyme activities positively correlated to TAS (r ¼ 0.60, Po0.001).

Objective: Routine high-dose Fe supplementation in non-anaemic pregnant women may induce oxidative stress and eventually affect birth outcomes. The aim of the present study was to measure oxidative stress markers in pregnant women with low/normal and high Hb values in trimester 1 (Hb 1 ) and to relate these to birth weight. Design: A cross-sectional study where selected oxidative stress markers were analysed in both maternal (trimester 1; T1) and cord blood samples and correlated with birth weight. Setting: A tertiary hospital in urban South India. Subjects: One hundred women were chosen based on their Hb 1 values (forty women with low/normal Hb 1 (<110 g/l) and sixty women with high Hb 1 (≥120 g/l)). Results: In T1, women with high Hb 1 values were found to have lower paraoxonase-1 (PON-1) activity (424·7 (SD 163·7) v. 532·9 (SD 144·7) pmol p-nitrophenol formed/min per ml plasma, P = 0·002) and higher lipid peroxides compared with women with low/normal Hb 1 . Routine supplementation of Fe to these women resulted in persistent lower PON-1 activity in cord blood (P = 0·02) and directionally lower (P = 0·142) birth weights. Furthermore, women with high Hb 1 who delivered low-birth-weight babies were observed to have lowest PON-1 activity in T1. No changes were observed in other markers (myeloperoxidase activity and total antioxidant levels). Conclusions: Routine Fe supplementation in pregnant women with high Hb 1 associated with increased oxidative stress, as reflected by low PON-1 activity in T1, could potentially lead to deleterious effects on birth weight.
