Maternal Eomesodermin regulates zygotic nodal gene expression for mesendoderm induction in zebrafish embryos

Abstract: Development of animal embryos before zygotic genome activation at the midblastula transition (MBT) is essentially supported by egg-derived maternal products. Nodal proteins are crucial signals for mesoderm and endoderm induction after the MBT. It remains unclear which maternal factors activate zygotic expression of nodal genes in the ventrolateral blastodermal margin of the zebrafish blastulas. In this study, we show that loss of maternal Eomesodermin a (Eomesa), a T-box transcription factor, impairs zygotic e… Show more

“…In zebrafish, perturbation analysis reveals that Eomesa is required for the activation of sox17 , as well as the two nodal ligands squint and cyclops [45]. ChIP analyses confirm that Eomesa physically binds to the squint locus [46,47]. Eomesa also binds regulatory regions near sox17 , and additional binding was observed for mixl1 , foxa , foxa3 , vent , and gsc [46].…”

“…Eomes and FoxH1 cooperate in controlling BMP/Nodal target genes together with Smads (Arnold et al, 2008;Bjornson et al, 2005;Miyazono et al, 2018;Slagle et al, 2011). Eomes, FoxH1, and Mixl1 have been implicated separately or in pairwise combinations in mesendoderm development Du et al, 2012;Henry and Melton, 1998;Kikuchi et al, 2000;Slagle et al, 2011;Xu et al, 2014). In particular, Eomes and MixL1 have been found to form a tripartite complex with Gata5 in early endoderm determination (Bjornson et al, 2005).…”
