Maternal Apolipoprotein B100 Serum Levels are Diminished in Pregnancies with Intrauterine Growth Restriction and Differentiate from Controls

Abstract: These results indicate the potential of liquid chromatography-multiple-reaction-monitoring mass spectrometry to become of clinical importance in the future for intrauterine growth restriction risk assessment based on maternal apolipoprotein B100 serum levels.

“…The aqueous phase of each peptide mixture was collected using a gel loading tip and reduced in volume to final volume of around 100 µL, each using a centrifugal evaporator (Speedvac RVC 2-25 CD plus, Martin Christ GmbH, Osterode am Harz, Germany). The peptide concentrations were measured using the Qubit assay as described [ 44 ], followed by desalting with OASIS cartridges [ 45 ]. Peptide concentrations of desalted peptide solutions were measured using the Qubit assay [ 44 ].…”

“…Equilibration and washing solutions consisted of 1% HCOOH in deionized H 2 O; two times 10 l were used for each step. The affinity-bound peptides were eluted into 5 l of 1% HCOOH in H 2 O : 1% HCOOH in ACN (50:50, v/v) (46,48). Next, all nine desalted portions of the tryptic peptide-containing solutions were pooled to obtain a total volume of 45 l. This solution was aliquoted into 10 l volumes, shock-frozen and kept at Ϫ20°C until either mass spectrometric analysis or immune complex formation were performed.…”

“…In contrast, women carrying females versus males may present greater stimulated cytokine production throughout pregnancy 17 . Furthermore, the maternal plasma proteome has been found to change as a function of the length of gestation 15,21 and mothers diagnosed with fetuses small for gestational age 29 and intrauterine growth restriction 30 have shown altered proteome profile 29,30 . In addition, placental proteins have been found to be associated to onset of labour and mode of delivery 31 .…”

“…In addition, placental proteins have been found to be associated to onset of labour and mode of delivery 31 . Taken together, both maternal factors, fetal sex and birth outcomes have been associated with certain proteins during pregnancy or delivery and deviations from a healthy pregnancy/delivery might be expressed in inflammation proteins related to these determinants 26,27,[29][30][31][32][33][34][35] .…”
