Mast Cell Proteoglycans

Abstract: Mast cells are versatile effector cells of the immune system, contributing to both innate and adaptive immunity toward pathogens but also having profound detrimental activities in the context of inflammatory disease. A hallmark morphological feature of mast cells is their large content of cytoplasmic secretory granules, filled with numerous secretory compounds, including highly negatively charged heparin or chondroitin sulfate proteoglycans of serglycin type. These anionic proteoglycans provide the basis for t… Show more

“…Such a stimulation might be comparable to that induced by soluble anti-IgE. As b-hexosaminidase is not associated with the serglycin matrix 42 , it immediately diffuses in the extracellular medium independently of the stimulation condition. As a consequence, the amount of b-hexosaminidase measured in the cell supernatant is similar following both localized and non-polarized stimulation.…”

“…Since toluidine blue is a cationic dye, it exhibits the capacity to selectively integrate with the glycosaminoglycan-protein complex (highly ionized) 11 . A hallmark feature of mast cells from all species is their striking metachromatic staining with various cationic dyes such as toluidine.…”

“…A role for proteoglycans in the homeostasis and storage of proteases and amines in secretory granules has been well studied in mast cells (Rönnberg et al 2012, Wernersson & Pejler 2014. For the exocrine pancreas, however, studies showing the presence of proteoglycans in ZGs have so far been descriptive only.…”

“…The latter enclose sulfated proteoglycans, which have been identified in ZG and in the fluid of the pancreatic ductal system (Berg & Young 1971, Reggio & Palade 1978, Tartakoff et al 1975. The negatively charged proteoglycans in secretory granules of hematopoietic cells and mast cells are involved in the binding of small positively charged molecules, such as amines and proteases, and have therefore been considered to promote the efficient storage of secretory products (Rönnberg et al 2012, Wernersson & Pejler 2014. Similarly, proteoglycans within ZGs have been proposed to support ZG biogenesis and to concentrate at the ZG membrane (Kleene et al 1999, Scheele et al 1994, Schmidt et al 2000, Schrader 2004), but their role in ZG formation is unclear.…”
