Mass spectrometry and the amyloid problem—How far can we go in the gas phase?

Abstract: A number of proteins are capable of converting from their soluble, monomeric form into highlyordered, insoluble aggregates known as amyloid fibrils. In vivo, these fibrils, which accumulate in organs and tissues, are associated with a wide range of amyloid diseases for which there are currently no therapeutic solutions. The molecular details of the pathway from native monomer through oligomeric intermediates to the final amyloid fibril remain a challenging enigma. Over the past few years, mass spectrometry has… Show more

“…To gain more insight into soluble oligomeric species invisible to NMR, IM-MS experiments were performed on samples with varying molar ratios of EGCG under conditions designed to probe their solution stoichiometry and structure (35,36,39). In the absence of EGCG, Aβ (10 μM) existed primarily as a mixture of monomers and dimers (Fig.…”

“…This correlation greatly expands the scope of gas phase structural investigations such as ion mobility measurements [22][23][24], gas phase hydrogen exchange [25], and optical spectroscopy [26]. In addition, a host of ion activation methods are available for the dissociation of protein assemblies [27][28][29][30][31][32].…”
