Mass spectrometric quantification of MIF-1 in mouse brain by multiple reaction monitoring

Abstract: MIF-1 (Pro-Leu-Gly-NH2) has potent therapeutic effects in depression and Parkinson’s disease, but its CNS sites of production are not yet clear. In this study, the concentration of MIF-1 in different brain regions was measured by the multiple reaction monitoring technique on a 4000 QTRAP mass spectrometer. The limit of quantification was 300 fg of MIF-1, and limit of detection was 60 fg. The low molecular weight fractions of tissue homogenates from different regions of mouse brain were analyzed. The concentrat… Show more

“…As originally identified by Kastin ( Kheterpal et al, 2009 ), the N-terminal fragment of oxytocin, proline-leucine-glycine-NH 2 (OT 7-9), is also known as melanocyte-inhibiting factor-1 (MIF-1). Among the functions of MIF-1 is the regulation of pain through interactions with the opioid receptors ( Khan et al, 2010 ).…”

“…Methods for protein labeling, electrophoresis, imaging, and analysis have been described in detail previously. [14][15][16] Briefly, protein samples were covalently labeled at lysines with spectrally resolvable CyDye fluorophores (Cy2, Cy3, and Cy5). The internal control pooled sample consisting of equal amounts of protein from each sample was labeled with Cy2 and included for each gel.…”

“…In recent years, this method has been modified and applied to measure concentrations of multiple NPs in different chemical environments. Kheterpal et al (76) employed stable isotopes 13 C and 15 N on the leucine residue to generate a standard curve for MIF-1 (Pro-Leu-Gly-NH 2 ), which has potent therapeutic effects in depression and Parkinson’s disease, thereby facilitating measurement of its actual concentration in the mouse brain.…”
