Mass spectrometric analysis of chondroitin sulfate-linked peptides

Abstract: Chondroitin sulfate proteoglycans (CSPGs) are extracellular matrix components composed of linear glycosaminoglycan (GAG) side chains attached to a core protein. CSPGs play a vital role in neurodevelopment, signal transduction, cellular proliferation and differentiation and tumor metastasis through interaction with growth factors and signaling proteins. These pleiotropic functions of proteoglycans are regulated spatiotemporally by the GAG chains attached to the core protein. There are over 70 chondroitin sulfat… Show more

“…Interestingly, S54 is conserved across placental mam-mals (among species sampled) while S57 is restricted to primates [6,7]. S54 can also be linked to chondroitin sulfate, a glycosaminoglycan polysaccharide of the extracellular matrix, identifying PGRMC1 as a proteoglycan core protein [463]. In the folded PGRMC1 protein, these residues are proximal to both the 47-49 RGD motif and the 62-66 SH3target motif.…”

“…Proteomic database searches were designed to include variable modifications of the residual 6-mer structure (993.2808 u) for [HexA(-H 2 O)GalNAcGlcAGalGalXyl- O -] with 0, 1, or 2 sulfates or phosphates attached, and all hits were manually validated and interpreted with regard to peptide sequence, glycan structure, and precursor mass. In total, the glycoproteomic analysis identified 21 previously established CSPGs as well as five novel CSPGs ( 2 , 31 ), including membrane-associated progesterone receptor component 2 (O15173), transmembrane protein 119 (Q4V9L6), low-affinity immunoglobulin epsilon Fc receptor (P06734), polypeptide N-acetylgalactosaminyltransferase 2 (Q10471) and matrix extracellular phosphoglycoprotein (Q9NQ76) ( supplemental Fig. S1 , A – E ).…”

“…In the latter, PGRMC1 was associated with the exosome pathway. PGRMC1 was also recently detected as a chondroitin sulfate-linked protein in urine [365]. The glycosaminoglycan was linked via S54 of PGRMC1, and therefore indicates a type 2 topology.…”
