Mass and Information Feedbacks through Receptor Endocytosis Govern Insulin Signaling as Revealed Using a Parameter-free Modeling Framework

Abstract: Insulin and other hormones control target cells through a network of signal-mediating molecules. Such networks are extremely complex due to multiple feedback loops in combination with redundancy, shared signal mediators, and cross-talk between signal pathways. We present a novel framework that integrates experimental work and mathematical modeling to quantitatively characterize the role and relation between coexisting submechanisms in complex signaling networks. The approach is independent of knowing or unique… Show more

“…As the models are often high-dimensional and contain many unknown parameters, parameter estimation does in general not provide a unique result, especially considering the limited amount of data. One common way to deal with that property is to obtain a sample of potential * E-mail: {vehlow, heinrich, weiskopf}@visus.uni-stuttgart.de † E-mail: {hasenauer, kramer, radde, allgower}@ist.uni-stuttgart.de parameters that provide a reasonable fit of the data [8,21,37]. The analysis of the model uncertainties, encoded within these samples, is essential to draw grounded conclusions about the systems' behavior.…”

“…Modes of administration are identified in a dialectical method depending on the condition of the patients. From a network pharmacology perspective, chronic diseases, especially T2DM, are a multi-mechanism, multi-gene disease, involving numerous metabolic pathways, can apply the concepts of compound compatibility in TCM [21]. For example, a drug intervention target network is often a subset of the network of biomolecules involved in the disease [22].…”

“…We use a modified version of the simannealingSBAO function that initiates multiple simplexes that are situated far away from each other in the space of parameters. With the multiple simplexes we are able to find multiple areas in the space of parameters -areas which potentially include additional acceptable parameters [13,23].…”
