Marshall R. Urist and the discovery of bone morphogenetic proteins

Abstract: Over the last 40 years International Orthopaedics has published a series of articles on bone morphogenetic proteins (BMPs) covering topics from basic research to clinical applications. This includes also work submitted from the Laboratory for Mineralized Tissues of the School of Medicine University of Zagreb. Accordingly, we felt obliged to give a short summary of Dr. Urist's life and work as our gratitude to his discovery that demineralized bone matrix (DBM) activity induces bone when implanted ectopically in… Show more

“…In 1965, Marshall Urist demonstrated for the first time that demineralized bone has capacity to induce new bone when implanted at ectopic sites, which he described as Bbone formation by auto induction^ [12,13]. Reddi has shown that implantation of demineralized bone matrix induces a cascade of cellular events involving mesenchymal cell recruitment, proliferation, and differentiation into cartilage-forming cells.…”

“…The preparation process included debridement of adherent soft tissues and removal of blood and lipids. Then, DBM was prepared by modified Urist method [ 64 ] and lyophilized for later use. In primary grinding, freeze‐dried DBM was grinded with zirconia balls with a diameter of 1.5 cm in grinding mill (CryoMill, Tetsch, Germany) 5 min at the speed of 30 per min.…”

“…He named this substance, the bone inductive principle, bone morphogenetic protein (BMP) and initiated an extensive and difficult search for these active protein molecules [2]. More than two decades later, advances in molecular biology by Dr. John Wozney's research team at the Genetic Institute described the first isolated extraction and recombinant form of BMP-2 in 1988 [3,4]. BMP-2 and BMP-7 have been shown to have the most bone forming potential out of the 15 BMPs identified and studied to date [5].…”
