Mapping the binding domains of the αIIb subunit

Abstract: a IIb b 3 , a member of the integrin family of adhesive protein receptors, is the most abundant glycoprotein on platelet plasma-membranes and binds to adhesive proteins via the recognition of short amino acid sequences, for example the ubiquitous RGD motif. However, elucidation of the ligandbinding domains of the receptor remains controversial, mainly owing to the fact that integrins are conformationally labile during purification and storage. In this study, a detailed mapping of the extracellular region of th… Show more

“…Flow cytometry results are presented as the mean fluorescence intensity of the activated sample minus the mean fluorescence intensity of the unactivated sample as previously described. 9 Platelet activation in vivo was also studied by measuring the plasma levels of soluble CD40L (sCD40L) with a commercially available ELISA kit (Bender Medsystems). Serum lipid levels and biochemical markers of liver and muscle function were determined with an automatic analyzer (Olympus AU560).…”

“…PAC-1 is a monoclonal antibody that binds to the activated form of the integrin receptor αIIb/β3 (30). The activation of this integrin leads to its conformational change and the recognition of various ligands, primarily fibrinogen, resulting in platelet aggregation and further activation through αIIb/β3-mediated outside-in signaling (26).…”
