Mapping of A1 conferring resistance to the aphid Amphorophora idaei and dw (dwarfing habit) in red raspberry (Rubus idaeus L.) using AFLP and microsatellite markers

Abstract: Background: Raspberry breeding programmes worldwide aim to produce improved cultivars to satisfy market demands and within these programmes there are many targets, including increased fruit quality, yield and season, and improved pest and disease resistance and plant habit. The large raspberry aphid, Amphorophora idaei, transmits four viruses and vector resistance is an objective in raspberry breeding. The development of molecular tools that discriminate between aphid resistance genes from different sources wi… Show more

“…Several genetic maps have been constructed for red raspberry using a variety of marker types, including amplified fragment length polymorphic (AFLP), genomic simple sequence repeat (SSR), expressed sequence tag-SSR (EST-SSR), and gene-based markers (Graham et al 2004(Graham et al , 2006Sargent et al 2007;Woodhead et al 2008Woodhead et al , 2010. The first published genetic linkage map of Rubus was developed using AFLP and SSR markers in progeny from an intraspecific cross between elite cultivars of red raspberry 'Latham' and 'Glen Moy'.…”

“…Base chromosome numbers (x) are indicated. The phylogeny illustrates the relative distances between the sub-families Rosoideae and Spiraeoideae, and between the genera within each sub-family A second genetic linkage map that consists of seven LG constructed of 95 AFLP, 22 SSR, and 2 gene-based markers, was developed from another intra-specific cross between elite cultivars of red raspberry 'Malling Jewel' and 'Malling Orion' (Sargent et al 2007). While this map is considered saturated, the high number of AFLP markers limits marker transferability.…”

“…Transmission of these viruses requires as little as a two minute feeding period on a healthy host plant (McMenemy et al 2009). Over the last 40-50 years, there have been extensive efforts to control this aphid by breeding resistant varieties (Sargent et al 2007) but these resistant varieties have applied strong selection pressure over many years. This, combined with the short generation time of the aphid, has caused the gradual evolution of aphid biotypes which are able to overcome plant resistance (McMenemy et al 2009).…”

“…Therefore, different molecular markers are very useful to distinguish between genotypes and for study of genetic diversity or relatedness. Markers such as RAPD (Graham and McNicoll, 1995), AFLP (Graham et al, 2004;Sargent et al, 2007;Miyashita et al, 2015), and SSR (Graham et al, 2002;2004;Badjakov et al, 2005;Stafne et al, 2005;Lopes et al, 2006;Sargent et al, 2007) have been applied also for Rubus species. Combined germplasm characterisation including both phenotypical and DNA-based methods is the most effective approach to compensate the shortcomings of each method.…”
