Mapping macrophage polarization over the myocardial infarction time continuum

Abstract: In response to myocardial infarction (MI), cardiac macrophages regulate inflammation and scar formation. We hypothesized that macrophages undergo polarization state changes over the MI time course and assessed macrophage polarization transcriptomic signatures over the first week of MI. C57BL/6 J male mice (3–6 months old) were subjected to permanent coronary artery ligation to induce MI, and macrophages were isolated from the infarct region at days 1, 3, and 7 post-MI. Day 0, no MI resident cardiac macrophages… Show more

“…Reduced neutrophil numbers observed in female hearts after MI also protects from collateral damage incurred by neutrophil-derived mediators. While both neutrophils and macrophages use LXR/RXR signaling and are equally responsible for adverse post-MI remodeling [58, 67, 78], the neutrophil difference was prominent. Future studies that investigate the role of other immune cells, including macrophages and T and B lymphocytes, are needed to fully understand all sex differences in cardiovascular disease pathology.…”

“…Groups were randomly assigned prior to the surgeries by one investigator (KYDP) and another investigator (YM) performed the majority of surgeries. To reduce animal use as recommended by the National Centre for the Replacement, Refinement, and Reduction of Animals in Research [46], the fibroblasts used in this study were obtained from a subset of the same mice used for macrophage isolation [36]. …”

“…To produce permanent MI, mice underwent coronary artery ligation surgery as described previously and according to the Guidelines for Experimental Models of Ischemia and Infarction [10, 21, 24, 31, 36]. Mice were anesthetized with 2% isoflurane, intubated, and ventilated.…”

“…Mouton et al analyzed macrophage physiology over the duration of MI, determining that macrophages isolated from mice 3 days post-MI exhibited an enriched phagocytic and proliferative phenotype [129]. A depletion of MertK, a specific marker of phagocytosis, correlates with prolonged inflammations due to an insufficient clearance of apoptotic cells [196].…”
