MAPKs and their relevance to arthritis and inflammation

Abstract: Rheumatoid arthritis (RA) is a chronic autoimmune disease in which imbalances in pro- and anti-inflammatory cytokines promote the induction of autoimmunity, inflammation and joint destruction. The importance of inflammatory cytokines in the pathogenesis of RA has been underscored by the success of biologics that act to block the effects of cytokines, such as tumour necrosis factor-alpha, interleukin (IL)-1 or IL-6, in treating disease. Mitogen-activated protein kinases (MAPKs) have been implicated as playing k… Show more

“…MAPKs are associated with TNF receptor-family signaling, chemokine-and cytokine-induced signaling events, and Toll-like receptor activation (40,41). In turn, MAPK signaling leads to the expression of proinflammatory cytokines and chemokines, such as TNF␣, IL-1, IL-6, IL-12, and IL-23 (41).…”

In turn, MAPK signaling leads to the expression of proinflammatory cytokines and chemokines, such as TNF␣, IL-1, IL-6, IL-12, and IL-23 (41). Notably, JNK activation early in RA, prior to evidence of fulfillment of the American College of Rheumatology/European League Against Rheumatism criteria (42), is predictive of subsequent erosions (43).…”

“…29 The extracellular signal-regulated kinases (ERKs) and p38 MAPK are two major classes of MAPKs that play important roles in transducing inflammatory signals. 30,31 To determine whether these two pathways are involved in the upregulation of IL-6 and IL-8 in ARPE-19 cells, we examined the activation of signaling molecules by western blotting with phospho-specific antibodies.…”
