Manipulation of the Anoxic Metabolism in Escherichia coli by ArcB Deletion Variants in the ArcBA Two-Component System

Abstract: Bioprocesses conducted under conditions with restricted O 2 supply are increasingly exploited for the synthesis of reduced biochemicals using different biocatalysts. The model facultative anaerobe Escherichia coli has elaborate sensing and signal transduction mechanisms for redox control in response to the availability of O 2 and other electron acceptors. The ArcBA two-component system consists of ArcB, a membrane-associated sensor kinase, and ArcA, the cognate response regulator. The tripartite hybrid kinase … Show more

“…Indeed, the activity of several dehydrogenases in cellular extracts was previously shown to be increased in an arcA mutant. However, the fact that the NADH/NAD + ratio is altered in an arcB strain [70] may be explained by the additional roles of ArcB beyond regulating ArcA [39], [71].…”

“…This strategy has been used to manipulate bacterial metabolism to enhance the synthesis of different bioproducts, especially reduced metabolites (13). On the other hand, concern about the costs of energy used for aeration in bioprocesses has renewed attention to the regulation of aerobic and anaerobic bacterial metabolism as a means to achieve the sustainable synthesis of a variety of bioproducts under these conditions (14).…”

“…Such metabolic engineering strategies, recently reviewed by Matsumoto et al . (), not only encompass the manipulation of target structural genes but also global regulators of metabolism, such as the ArcAB two‐component system (Bidart et al ., ). These platform E .…”
