Manipulating Intratumoral Fenton Chemistry for Enhanced Chemodynamic and Chemodynamic‐Synergized Multimodal Therapy

Abstract: Chemodynamic therapy (CDT) uses the tumor microenvironment‐assisted intratumoral Fenton reaction for generating highly toxic hydroxyl free radicals (•OH) to achieve selective tumor treatment. However, the limited intratumoral Fenton reaction efficiency restricts the therapeutic efficacy of CDT. Recent years have witnessed the impressive development of various strategies to increase the efficiency of intratumoral Fenton reaction. The introduction of these reinforcement strategies can dramatically improve the tr… Show more

“…With the development of nanomedicine, CDT has received increasing attention due to its specific activation mechanism in the TME [9,10]. CDT relies on the Fenton/Fentonlike activity between the less active hydrogen peroxide (H 2 O 2 ) and Fe, Cu, Co or Mnbased reagents to generate the most cytotoxic reactive oxygen species (ROS), hydroxyl radicals (OH), inducing apoptosis and necrosis in cancer cells [11][12][13].…”

“…The fluorescence intensity of cellular Rho was quantified ( Figure 2(B) ), showing that the cellular uptake of the PP@Mn NPs increased within 8 h. The Mn 2+ in tumor cells can catalyze H 2 O 2 to •OH through the Fenton-like reaction (Liu et al., 2020 ; Ou et al., 2021 ). Moreover, tumor PTT can elevate the •OH level while improving the efficiency of the Fenton-like reactions (Liu et al., 2020 ; Zhou et al., 2021 ). Therefore, the PP@Mn NPs were designed and applied to the combination therapy of CDT and PTT.…”

“…In Europe, Nanotherm ® , which is used as a nano agent of magnetic hyperthermia for brain gliomas, has been approved for clinical use [ 13 ]. MIONs also have the ability to kill cancer cells by producing reactive oxygen species (ROS) through the Fenton reaction catalyzed by Fe 2+ , which is known as chemodynamic therapy (CDT) [ 14 ]. Additionally, MIONs can integrate with other materials for multi–modality therapy or diagnostics ( Figure 1 ).…”
