Manganese uptake, mediated by SloABC and MntH, is essential for the fitness ofStreptococcus mutans

Abstract: Early epidemiological studies implicated manganese (Mn) as a possible caries-promoting agent while laboratory studies have indicated that manganese stimulates the expression of virulence-related factors in the dental pathogen Streptococcus mutans. To better understand the importance of manganese homeostasis to S. mutans pathophysiology, we first used RNA sequencing to obtain the global transcriptional profile of S. mutans UA159 grown under Mn-restricted conditions. Among the most highly expressed genes were th… Show more

“…Total RNA was isolated from E. faecalis OG1RF cells grown to mid-log phase in FMC[+Fe] or FMC[-Fe] or grown to mid-log phase in FMC[+Fe] and transferred to FMC[-Fe] following the methods described elsewhere (80). The RNA was precipitated with ice-cold isopropanol and 3 M sodium acetate (pH 5) at 4°C before RNA pellets were suspended in nuclease-free H 2 O and treated with DNase I (Ambion) for 30 min at 37°C.…”
