a b s t r a c tIn the marine environment, manganese represents a potential emerging contaminant, resulting from an increased production of manganese-containing compounds. In earlier reports we found that the exposure of Paracentrotus lividus sea urchin embryos to manganese produced phenotypes with no skeleton. In addition, manganese interfered with calcium uptake, perturbed extracellular signal-regulated kinase (ERK) signaling, affected the expression of skeletogenic genes, and caused an increase of the hsc70 and hsc60 protein levels. Here, we extended our studies focusing on the temporal activation of the p38 mitogen-activated protein kinase (p38 MAPK) and the proteolytic activity of metalloproteinases (MMPs). We found that manganese affects the stage-dependent dynamics of p38 MAPK activation and inhibits the total gelatin-auto-cleaving activity of MMPs, with the exclusion of the 90e85 kDa and 68e58 kDa MMPs, whose levels remain high all throughout development. Our findings correlate, for the first time to our knowledge, an altered activation pattern of the p38 MAPK with an aberrant MMP proteolytic activity in the sea urchin embryo.

Abstract:The current study was carried out for examining the heavy metal pollution profile of seven urban ponds in Chavara industrial area, south west coast of India. The study was conducted seasonally for a period of one year from January 2014 to December 2014. Results obtained from AAS analysis showed that toxic heavy metals such as copper (Cu), Manganese(Mn), zinc(Zn), Iron(Fe), lead(Pb), Cadmium(Cd), Nickel(Ni), Cobalt (Co), and Chromium(Cr) in the water samples were found to be higher than drinking water standards prescribed by WHO, BIS and FAO. This study is an attempt to compare the hydrological assessment of heavy metals in water of different ponds due to pollution from near industry-KMML, situates in Chavara, Kollam, India.

Natural matrices affect environmental DNA (eDNA) detections. Effects of matrix‐eluted compounds on the polymerase chain reaction (PCR) step have been the focus of most inhibition studies. Factors affecting eDNA detections in a typical laboratory workflow, i.e., DNA extraction and PCR steps, are mostly unknown. Here, we assessed the effect of four metal ions (Ca2+, Fe3+, Mn2+, Cu2+) present in marine sediments on DNA detectability for both the extraction and the PCR detection steps. A single metal ion and exogenous DNA were added to marine sediments treated chemically to remove inhibitors. Our results showed that natural concentrations of calcium, iron, and manganese ions in surface marine sediments can impede completely DNA detections. Alternatively, copper ions added to the matrix increased DNA detectability by 7.7%. We also observed bimodal inhibitory effects of calcium and iron ions on DNA detectability, suggesting that the extraction and the PCR steps are both affected. Our findings highlight new limitations of eDNA detections. Avenues to optimize eDNA detection protocols applicable to multiple matrices are discussed.
