Mancozeb impairs mitochondrial and bioenergetic activity in Drosophila melanogaster

Abstract: Mancozeb (MZ) is a broad-spectrum fungicide used worldwide in several crops. Neurological disorders in humans and animals have been associated with exposure to this compound by mechanisms still not fully understood. Drosophila melanogaster represents a reliable model in toxicological studies, presenting genetic and biochemical similarities with mammals. In this study, D. melanogaster flies were exposed for 15 days to MZ through the food (5 and 10 mg/mL). After that period, the efficiency of mitochondrial respi… Show more

“…Dithiocarbamate pes ticides like mancozeb (MCZ) are regarded as significant fungicides with applications worldwide by farmers from various countries [8]. Despite the wide application and acceptance of (MCZ) fungicides, it was found to be lethal and detrimental to off-target essential organismandhumanbeings.Mancozebwasfound to impart neurotoxicity, damaging mitochondria, andmyofibrilsplitting [9], thyroid disruption [10], changes in glutathione and alterations in essential metal homeos tasis [11], adverse effects on s teroid production [12], ovarian injury and apoptosis [13], induce alteration in the development of hypothalamic sys tems that are essential for maturation of neuroendocrine sys tem [14], disrupt mitochondria and bioenergetic activities in the body [15], affect male fertility [16] and induce behavioural deficit [17].Although(MCZ)iswidelyacceptedandused by farmers in many countries, these and many other reports have proven that the uncontrolled and excessiveapplicationof(MCZ)fungicideisacase for alarm. Therefore, there is an urgent and crucial need to devise a means through which the residues ofthesetoxicpes ticidescanbereducedordegraded in the environment.…”

“…Additionally, ETU is employed as an accelerator in rubber vulcanization [ 13 ]. As a mechanism of action, ETU disrupts protein synthesis and metabolism by inactivating amino acidic sulfhydryl groups in the protein structure of fungal pathogens [ 14 ]. ETU is produced as a contaminant or as degradation product when fungicides are stored in the presence of moisture and oxygen.…”
