This study was to investigate the effect of subcutaneous injection of the adipose stem cells (ASCs) with conditioned media (CM) in the treatment of acne vulgaris scar. We used Adult male New Zealand white rabbit ears as an animal model and induced acne formation by Kignman method. Adipose tissue was isolated and harvested from the scapula of rabbits, and ASCs were cultured and expanded until passage 1. There have four groups in our experiment, include phosphate buffered saline (PBS), ASCs with PBS (ASC ? PBS), CM, and ASCs with CM (ASC ? CM) group. This solution of 0.6 ml injected to subcutaneous in each group. ASC ? PBS and ASC ? CM groups were containing ASCs of 5.0 9 10 6 cells/ml. We analyzed the treatment of 4 groups to scar tissue after 2 and 4 weeks by hematoxylin and eosin stain, immunohistochemistry, and RNA expression level of tumor necrosis factor-a (TNF-a), interleukin-1a (IL-1a), and matrix metalloproteinase-2 (MMP-2). Also, the expression of keratin 16 (K16) was detected by western blot analysis. H&E stain showed that infiltration of inflammation cells was significantly reduced at 2 and 4 weeks, as well as re-epithelialization was improved in the ASC ? CM group. The ASC ? CM gourp was reduced both expression levels of TNF-a, IL-1a, and MMP-2 and K16 protein level. In conclusion, the ASCs with CM has a significant curative effect on acne vulgaris scar, more to the point, the CM has a key role on treatment. It could be applied to a therapeutic approach to regenerate to treat acne vulgaris scar.

Background: Acne is a chronic disorder that affects almost 80% of adolescents and young adults, causing psychological and emotional distress. However, the current treatments for acne are either ineffective or have many side effects. This study was designed to confirm and objectively quantify the effect of a new non-drug combined therapy on acne.Methods: This study innovatively utilized ultrasound, which enhanced the absorption of aloe vera gel, and soft mask to make a purely physical method without any drugs. In both the treatment group and control group, the number of papules/pustules and the area of hyperpigmented lesions were counted, and a smart mirror intelligent face system was used before and after the combined therapy. Alterations in the skin functional index were recorded and analyzed statistically.Results: In the treatment group, the combined therapy significantly reduced the number of papules and the area of hyperpigmented lesions and improved skin roughness and local blood circulation. In the control group, there was no obvious improvement over 2 months.Conclusion: This study suggests that the new non-drug combined therapy significantly improved acne, which provided experimental evidence and treatment guidance for patients with mild to severe acne, especially patients with moderate acne. This new therapy may possibly be an appropriate method for patients who seek topical treatments with mild side effects and low antibiotic resistance rates.

<p>Isotretinoin merupakan pilihan terapi oral yang paling efektif untuk kasus akne vulgaris derajat berat, tidak responsif terhadap terapi konvensional, akne dengan jaringan parut berat, dan akne kronis. Efek samping mukokutaneus paling sering dan tergantung dosis (dose-dependent), diduga berkaitan dengan berkurangnya produksi sebum, berkurangnya ketebalan stratum korneum, dan berubahnya fungsi barier epidermis. ATRA (all-transretinoic acid) dapat meningkatkan ekspresi TRAIL, yang merupakan protein ligan TNF-related menginduksi apoptosis. Vitamin E (-tokoferol) dapat menekan jumlah reseptor ligan apoptosis serta mengatur profil lipid epidermal. Beberapa penelitian menunjukkan vitamin E dapat mengurangi efek samping isotretinoin.</p><p>Isotretinoin is the most effective choice of oral therapy for severe acne, acne unresponsive to conventional therapy, severe acne scars, and chronic acne. Mucocutaneous side effects are the most frequently occurring and dose-dependent, thought to be related to reduced sebum production, reduced stratum corneum thickness, and epidermal barrier function. ATRA (all-trans retinoic acid) increases the expression of TRAIL, a TNF-related ligand protein that induces apoptosis. Vitamin E (-tocopherol) can suppress apoptotic ligand receptors and regulate lipid profiles. Several studies show the advantages of vitamin E in reducing side effects of isotretinoin.</p>
